Semaglutide (injectable) vs Orforglipron
Key distinction is injectable peptide GLP-1 versus daily oral non-peptide GLP-1; compare evidence maturity, formulation, label, efficacy and tolerability.

Semaglutide (injectable)
Orforglipron
Human evidence
Extensive Phase III evidence supports weight loss with regulated semaglutide. SELECT demonstrated cardiovascular benefit in a defined population, and STEP 9 showed improved knee-OA pain/function in adults with obesity.
Large Phase III trials demonstrate clinically meaningful weight loss and glycaemic efficacy with oral orforglipron.
Research focus
Obesity, weight management, cardiovascular outcomes, heart failure, osteoarthritis, type 2 diabetes
Obesity, weight management, type 2 diabetes, cardiometabolic risk
Regulatory status
Wegovy (semaglutide) is authorised in the EU for weight management in defined adults and adolescents; authorisation and dosing are product-, route-, indication- and jurisdiction-specific.
FDA approved Foundayo (orforglipron) on 1 April 2026 for chronic weight management in adults with obesity, or overweight with at least one weight-related comorbidity; approval is product/indication/jurisdiction specific.
Route / formulation
Evidence for licensed pre-filled injection pens cannot establish identity, concentration, sterility or equivalence of unverified semaglutide vials or non-authorised products.
Current labelled Foundayo tablet strengths/dose escalation must be kept separate from numeric doses used in earlier Phase III trial publications.
No single definitive all-population head-to-head outcome basis in current dataset Key distinction is injectable peptide GLP-1 versus daily oral non-peptide GLP-1; compare evidence maturity, formulation, label, efficacy and tolerability without cross-trial winner claims.
